Recombinant Protein

An antibody is a protein produced by plasma cells of the immune system that recognizes and neutralizes antigens such as bacteria, virus, and foreign proteins. An antibody is composed of two large heavy chains and two small light chains, which are linked by disulfide bonds. The antigen binding sites are found in the variable region (V region) and the constant region (C region) houses the Fc that directs the appropriate immune response upon antibody binding to its specific antigen. A single chain variable fragment (scFv) is a recombinant protein composed of a variable heavy chain (VH) and a variable light chain (VL) from antibody’s V region. Its molecular weight is ¼ ~ ½ of a full antibody while maintaining similar antigen binding specificity and affinity. Mammalian expression system is used for scFv production and variety scFv specific to different antigen can be synthesized. Recombinant protein technology enables proteins of interest to be produced in larger amounts from E. coli or mammalian cells. Our service includes custom recombinant scFv antibodies, custom recombinant scFv fusion proteins, custom recombinant proteins, and recombinant fusion proteins expressed in mammalian system (or E. coli).

Custom recombinant scFv antibody and scFv fusion protein

We provide a custom-designed recombinant scFv antibody through advanced tools for scFv/Fab expression in mammalian system. Also, we produce a recombinant scFv fusion protein composed of an scFv antibody and active protein domains through recombinant protein technology. Generation of a bi-functional fusion protein containing an scFv region along with a functional protein preserves the intrinsic activities of both domains. With the initial 100 ml culture, we estimate to produce approximately 1 mg antibody. Additional order that has been placed within the 6 months from the first order can be placed per mg protein. For each antibody, one-time cloning fee will be applied.

  1. Produce a customized scFv antibody or a fusion protein expression vector
  2. Expression of the scFv antibody or the fusion protein in 293 cells (optional CHO cells available)
  3. Purification of the scFv antibody or the fusion protein
  4. Identification by SDS-PAGE and Western Blot
  5. Ship to customer (additional shipping fee to outside of the U of U)

 

Manufacturing time: 6-8 weeks without customer-supplied vector or 4-6 weeks with customer-supplied vector.

We provide variety of custom recombinant proteins and fusion proteins in mammalian cells (E. coli service: contact director for more details). Stocked items

We provide a high quality recombinant green fluorescence protein (GFP) and an activity-verified anti-HER2 scFv antibody. GFP is expressed in E. coli system and the anti-HER2 scFv antibody is expressed in 293 cells. We will update the list of stocked items as we establish a library of scFv antibodies and other functional recombinant proteins. 100 μg minimum; bulk order available.
